1、醋曲片的最佳制备工艺与质量控制,硕士论文高脂血症导致的心血管疾病在近年来发病率越来越高。其造成血脉壁的淤 积,极易产生急性冠心病和动脉粥样硬化性心血管疾病。为了减少心血管疾病的 发生,降脂治疗和预防势在必行。近年来,来源天然的降脂产品备受消费者的关 注。 本研究制备的醋曲片由醋粉、红曲和芦荟组成,用于降血脂,其疗效确切且 具有较好的安全性。 制剂中醋粉和红曲以粉末入药,芦荟经醇提浓缩枯燥之后入药。首先对芦荟 的提取工艺进行研究,采用乙醇加热回流提取法,选取乙醇浓度、提取时间、液 料比为考察因素,以黄酮类化合物的吸光度值R1和浸膏得率R2为评价指 标,采用响应曲面法对芦荟中总黄酮提取工艺进行优化

2、,确定最佳提取工艺为: 65%乙醇作提取溶剂,液料比为 20:1,回流提取 4 h,对提取液中的乙醇进行回 收,浓缩物于 60 烘箱中枯燥。 醋曲片片芯的制备采用湿法制粒压片,通过单因素考察发现崩解剂用量、润 湿剂浓度和枯燥温度对片芯质量影响较大,因而以上述三种变量为考察因素,以 片芯的崩解时间、硬度、外观和压片前颗粒的休止角等指标的综合评分作为成型 工艺的评价指标,采用正交试验设计对崩解剂用量、润湿剂浓度和枯燥温度进行 优化,确定了最优片芯1000 片处方为:醋粉470 g,红曲200 g,芦 荟280 g,预胶化淀粉75 g,聚乙烯吡咯烷酮25 g,85%乙醇溶液适 量,滑石粉10 g。

3、醋曲片中含挥发性和易吸湿性成分,因而片剂外包薄膜衣。选取包衣温度、 包衣液供应速率和包衣增重三个变量为考察因素,以所得包衣片的合格率和崩解 时间作为评价指标,采用 Box-Behnken 试验设计方案,对选取的三个包衣工艺参 数进行挑选。确定了最佳包衣工艺为:取适量薄膜包衣预混剂于纯化水中溶解, 制得固含量为 12%的包衣液,包衣温度为 45 ,包衣液供应速率为 4 mL/min, 包衣增重为 3%. 本研究建立了醋曲片的质量标准:采用显微鉴别法和薄层色谱法对醋曲片中 的红曲进行定性鉴别,采用薄层色谱法对醋曲片中的芦荟进行定性鉴别,以上药 材均检出且特征明显。采用高效液相色谱法测定醋曲片中总洛

4、伐他汀的含量,经 方式方法学验证,各项指标均符合相关规定,本研究建立的定性、定量方式方法准确可靠。 本论文确立了醋曲片的最佳制备工艺,经中试试验验证,工艺稳定可行。醋 曲片中的洛伐他汀含量高,用于降血脂疗效显著,因而以总洛伐他汀作为质量控 制的指标成分。经影响因素试验、加速试验及长期试验考察,结果表示清楚,醋曲片 稳定性良好。 本文关键词语:醋曲片,制备工艺,质量标准,洛伐他汀,稳定性 Abstract The incidence rate of cardiovascular disease caused by hyperlipemia has been increasing in recen

5、t years. It causes blood vessel wall siltation, which is very easy to produce acute coronary heart disease and atherosclerotic cardiovascular disease. In order to reduce the incidence of cardiovascular disease, lipid-lowering treatment and prevention is imperative. In recent years, natural lipid-low

6、ering products have attracted much attention of consumers. The vinegar koji tablet prepared in this study is composed of vinegar powder, Monascus and aloe. It is effective and safe for reducing blood lipid. In the preparation, vinegar powder and Monascus powder were used as medicine, aloe was used a

7、s medicine after alcohol extraction, concentration and drying. Firstly, the extraction process of Aloe Vera was studied. The ethanol concentration, extraction time and liquid-solid ratio were selected as the factors, and the absorbance value R1 and extraction yield R2 of flavonoids were used as eval

8、uation indexes. The extraction process of total flavonoids in aloe was optimized by response surface methodology 65% ethanol was used as extraction solvent, the ratio of liquid to material was 20:1, reflux extraction for 4 h, the ethanol in the extract was recovered, and the concentrate was dried in

9、 an oven at 60 。 The preparation of vinegar starter tablet core adopts wet granulation and tablet pressing. Through single factor investigation, it is found that the amount of disintegrating agent, wetting agent concentration and drying temperature have great influence on the quality of the core. Th

10、erefore, the above three variables are taken as the investigation factors, and the comprehensive score of disintegration time, hardness, appearance and the angle of repose of granules before compression is taken as molding The optimal formulation of 1000 tablets was determined as follows: vinegar po

11、wder 470 g, Monascus 200 g, aloe 280 g, pre gelatinized starch 75 g, polyethylenepyrrolidone 25 g, 85% ethanol solution appropriate, talcum powder 10 g。 Vinegar starter tablets contain volatile and hygroscopic components, so the tablets are coated with film. Three variables including coating tempera

12、ture, coating solution supply rate and coating weight gain were selected as the evaluation factors. The qualified rate and disintegration time of the coated tablets were used as evaluation indexes. The three coating process parameters were selected by box Behnken design. The optimal coating process

13、was determined as follows: a suitable amount of film coating premix was dissolved in purified water, and the coating solution with solid content of 12% was prepared. The coating temperature was 45 , and the coating solution supply rate was 4 ml / min, The coating weight gain was 3%. In this study, t

14、he quality standard of vinegar koji tablets was established: the red koji in vinegar starter tablets was qualitatively identified by microscopic identification and thin layer chromatography, and the aloe in vinegar starter tablets was identified by TLC. High performance liquid chromatography was use

15、d to determine the content of total lovastatin in vinegar starter tablets. Through the verification of the methods, all the indexes were in line with the relevant provisions. The qualitative and quantitative methods established in this study were accurate and reliable. In this paper, the best prepar

16、ation technology of vinegar koji tablets was established, which was proved to be stable and feasible by pilot test. The content of lovastatin in vinegar Qu tablets is high, so the total lovastatin is used as the index of quality control. The results showed that the stability of vinegar starter table

18、机中混合 20 min 后制备软材,制粒,烘干,整粒;压片前的总混物具引湿性, 应注意生产时环境湿度控制在 78.85%下面。其次,对片芯成型工艺中三个主要 影响因素进行了考察,确定了颗粒枯燥温度为 60 ,最优片芯1000 片处方 为:醋粉470 g,红曲粉200 g,芦荟280 g,预胶化淀粉75 g, 聚乙烯吡咯烷酮25 g,85%乙醇溶液适量,滑石粉10 g。对片芯进 行了包衣工艺的考察,在包衣增重 3%,包衣温度 45 ,包衣液喷速 4 mL/min 条件下,醋曲片的包衣合格率和崩解时间均良好。 接着对醋曲片的质量控制标准进行了研究。对红曲进行显微特征的鉴别,可 以观察到红曲的各项生

19、物学特征;建立了醋曲片中红曲、芦荟的 TLC 鉴别方式方法, 结果表示清楚其操作简单,结果准确,重现性良好;建立了醋曲片中总洛伐他汀的 HPLC 含量测定方式方法,并进行方式方法学考察,结果表示清楚此方式方法专属性强、简便高效, 可用于总洛伐他汀的含量控制。根据 2021 版(中国药典有关片剂项下的质量 标准要求,对醋曲片的片重差异、脆碎度、崩解时间和总洛伐他汀含量等进行了 检查,结果表示清楚各指标均符合药典要求。 最后经影响因素试验、加速试验和长期试验考察,表示清楚醋曲片产品具有良好 的稳定性。醋曲片的制备和质量控制方式方法准确高效,可为醋曲片的大量生产提供 参考。 致 谢 岁月如梭,三年光
