1、第 1 页 共 24 页单体车间发酵岗位工艺操作规程1.目的及适用范围1.1 本规定的目的是严格工艺操作,提高工作质量,保证安全文明生产。1.2 本规定适用于发酵工序消毒、接种、移种、温度控制、压力通气量控制、冷冻、取样、加热放罐、输送的过程控制。2.相关文件:无3.发酵岗位使用专业术语3.1 延滞期:培养基接种以后,细胞并不立即生长繁殖,其细胞数在一定时期内无明显增加,这一阶段称为延滞期。延滞期是细胞在环境改变后表现出来的一个适应阶段,这是一个静态到动态的过程,须缓慢控制,即用低转速来控制好溶氧。3.2 对数生长期:细胞经过延滞期后适应了新环境,生理状态也较为活跃,细胞开始迅速繁殖,由于细胞

2、以分裂方式繁殖,细胞数目呈几何级数增加,故称对数生长期。进入对数生长期可提高转速满足菌种对氧的高呼吸要求。3.3 稳定期:随着细胞的生长繁殖,培养基中营养物质渐趋耗尽,而代谢产物的逐渐增多,以及菌生长引起周围环境条件(如PH值、温度等)的变化,使的细胞的繁殖速度逐渐降低,当细胞的繁殖速率与死亡速率相等时,细胞生长进入稳定期,细胞浓度达到最大。3.4 衰亡期:培养基中营养成分耗尽,代谢产物大量堆积,这时能够继续繁殖的细胞数越来越少直至为零,而死亡的细胞则越来越多,即活细胞数显著下降,故此时期称为衰亡期。3.5 球状菌:球状菌又可细分为单球菌、双球菌、四连球菌、八叠球菌、链球菌、葡萄球菌。3.6

3、杆状菌:第 2 页 共 24 页3.6.1 按菌的长短细分为长杆菌和短杆菌。3.6.2 按菌的粗细细分为粗杆菌和细杆菌。3.6.3 按菌的相连个数细分为单杆菌、双杆菌、链杆菌。3.7 螺旋状菌:螺旋状菌又可细分为弧菌和螺旋菌。注:芽孢菌(又称内生孢子)不具有分类学上的意义,其只是细菌(一般为杆菌,球菌也有)在生长过程中细胞内部产生了一些圆状或椭圆状的空心,以抵抗不良环境对其的影响。4.职责:4.1 认真执行本公司各项管理规定和规章制度。4.2 严格按单体操作规程和设备操作规程操作,严禁超温、超压,违章操作。4.3 对本岗所有设备安全负责,定期对设备进行维护保养,冬季做好管线、设备的防冻凝工作。

20、种口外盖,插上针头,随后调整放空阀,使其有适量的蒸汽排出,罐内压力稳定在0.150.25MPa之间,消毒半小时以上。5.1.1.5 种子罐消完后取下针头,旋上接种口外盖,关闭三路蒸汽及放空阀。打开移种出料一次阀和种子罐排污阀,将罐内液体压空后关闭移种出料一次阀和种子罐排污阀。5.1.1.6 微开放空阀,打开空气进气一次阀、二次阀对罐体进行保压操作。(种子罐长久不用或染芽孢菌时,要加入适量的甲醛闷罐。)5.1.2 种子罐实消种子罐经空消后,根据生产需求,可以上罐实消。5.1.2.1 加水:打开加料口,加入自来水110 升,依据所给定的标准加入各种无机盐、尿素、酵母膏等培养基,加料完成后关闭加料口

21、。5.1.2.3更换接种口橡皮塞。5.1.2.4打开夹套排污阀,检查其他所有阀门是否处于关闭状态,打开夹套蒸汽一次阀和二次阀。对罐体进行预热,待温度升至90后关闭夹套蒸汽一次阀和二次阀。5.1.2.5 打开放空阀,将种子罐内压力排掉,关闭放空阀。通三路蒸汽,压力上升到 0.09MPa后开始调整放空阀,使其有微量的排汽,调整三路蒸汽进汽阀,使罐内压力稳定在0.080.11MPa 之间,温度上升至118后开始计时,消毒 20 分钟后,关闭三路蒸汽阀。开空气进气一次阀、二次阀,通气量尽可能小,调整放空阀,使罐内压力在0.1MPa左右,关闭底部排污阀。5.1.2.6 打开冷却循环水回水阀,打开冷却循环

29、冷却循环水进水阀。关冷却循环水回水阀,等待接种。5.1.3 种子罐的接种本操作由菌种室操作人员负责,本岗人员配合操作。5.1.4 种子罐的培养5.1.4.1 温度控制:2815.1.4.2 由于随着微生物的增殖和发酵会发热、搅拌产热等,为使罐内温度始终保持最佳,需对罐内进行温度控制。5.1.4.3打开种子罐 28 度水回水阀,打开28 度进水阀,检查种子罐排污阀是否关闭,检查种子罐 20 度水阀门是否关闭,检查夹套蒸汽阀是否关闭。打开 28 度水泵入口阀门,检查28 度水泵出口阀门是否关闭,开启28 度水泵,打开 28 度水泵出口阀门,使正在培养的种子罐温度控制在281,关闭种子罐 28 度水

30、进水阀和回水阀。5.1.4.4 种子罐降温:关闭种子罐28 度水进水阀和回水阀,打开种子罐20 度水回水阀;打开20 度水进水阀,进行降温,温度降到29-30 度,降温操作完毕。关闭种子罐20 度水回水阀;关闭20 度水进水阀。打开种子罐 28 度水回水阀和进水阀。5.1.4.5种子罐加热:关闭种子罐28 度水进水阀和回水阀,打开排污阀,依次打开夹套蒸汽一次阀、缓慢打开夹套蒸汽二次阀,对罐体进行加热。根据蒸汽压力控制每次加热时间,每次加热时间不应超过20 秒,关闭蒸汽二次阀,关闭蒸汽一次阀。温度恢复至正常范围后(时间为加热后510分钟左右),关闭夹套排污阀,打开种子罐28 度水回水阀和进水阀。
