6、1)为适应当时畜牧业生产发展的需要,某兽医实验室成立于19_年。近些年,受财政预算减少等因素影响,政府采取兽医实验室商业化、私有化等措施,如为养殖户和利益相关方提供有偿兽医实验室检测服务,以解决当前的问题。但兽医实验室也面临着科研人才流失、仪器设备老化和日益复杂的动物疫病流行形势等诸多挑战。为掌握该兽医实验室血凝抑制试验(HI )的检测能力,由6名具备检测资质的技术人员同时进行HI 抗体检测,笔者对试验结果进行了比对分析p 。1材料与方法1.1试验材料新城疫标准阳性血清(批号:2/12),新城疫标准阴性血清(批号:06/15A ),新城疫标准检测抗原(批号:080920_),均由意大利IZSA
7、M 实验室提供。1.2试验方法将鸡新城疫标准阴性、阳性血清分别进行倍比稀释后分装,共制成52份盲样,其中8份为阳性,即HI 效价116,结果(即参考值)记为N log2。6名检测技术员参照OIE 陆生动物诊断试验与疫苗手册20_第2.3.14章,对这52份盲样进行鸡新城疫病毒HI 抗体检测,检测结果记为n log2。1.3统计方法检测值(n )和参考值(N )均按4为阳性,反之为阴性进行定性判断。应用Epi Info 7对结果进行22表格统计(图1),计算阳性一致性(a a+c )、阴性一致性(d b+d ),用Episcope 2.0计算符合率(Observed proportion of
8、agreement )(a+d a+b+c+d),再用 Episcope 2.0软件进行试验一致性(Agreement )分析p ,计算Ka 值,以比较HI 定性试验的符合情况。参照经典文献进行一致性强度判断,即Ka 值 0.80,高度;0.811.00,极强1。用SPSS 20和E_cel 软件对检测值和参考值的差值进行统计描述和频数分析p ,统计差值的极大值、极小值、均值、标准差、方差、频数和直方图,分析p 定量结果的总体分布情况。2结果与分析p 2.1定性结果一致性将6名检测技术员的312个检测值与参考值的定性结果全部进行相关统计,结果检测阳性42份,阳性率13.5,低于参考值15.4的阳性率(字2=148.85,P 结果的一致性为68.8(33/48),阴性结果一致性为96.6作者简介刘华(1982-),男,安徽宁国人,硕士,兽医师,从事动物疫病防控与兽医流行病学研究工作。收稿日期20_-07-13图1检测值与参考值阴阳性结果对照参考值(N )检测值(n )230第 3 页 共 3 页
